The United States houses only five percent of the world’s population but over 20% of its prison population. There has been a dramatic increase in carceral numbers over the last several decades with much of this population being people with mental illness designations. Many scholars attribute this phenomenon to the process of deinstitutionalization, in which mental health institutions in the U.S. were shut down in the 1950s and ‘60s. However, disability scholar Liat Ben-Moshe argues that this is a dangerous oversimplification that fails to credit the deinstitutionalization movement as an abolitionist movement and to take into account shifting demographics between institutions and prisons/jails. This study considers how mass incarceration in the U.S. stems from a trend of isolating and punishing BIPOC and people with disabilities at disproportionate rates as it explores lived experiences at the intersection of mental health and incarceration. Findings inform an abolitionist agenda by highlighting the near impossibility of rehabilitation and treatment in an inherently traumatizing space.

Molecular engineering is an emerging field that aims to create functional devices for modular purposes, particularly bottom-up design of nano-assemblies using mechanical and chemical methods to perform complex tasks. In this study, we present a novel method for constructing an RNA clamp using circularized RNA and a broccoli aptamer for fluorescence sensing. By designing a circular RNA with the broccoli aptamer and a complementary DNA strand, we created a molecular clamp that can stabilize the aptamer. The broccoli aptamer displays enhanced fluorescence when bound to its ligand, DFHBI-1T. Upon induction with this small molecule, the clamp can exhibit or destroy fluorescence. We demonstrated that we could control the fluorescence of the RNA clamp by introducing different complementary DNA strands, which regulate the level of fluorescence. Additionally, we designed allosteric control by introducing new DNA strands, making the system reversible. We explored the use of mechanical tension to regulate RNA function by attaching a spring-like activity through the RNA clamp to two points on the RNA surface. By adjusting the stiffness of the spring, we could control the tension between the two points and induce reversible conformational changes, effectively turning RNA function on and off. Our approach offers a simple and versatile method for creating RNA clamps with various applications, including RNA detection, regulation, and future nanodevice design. Our findings highlight the crucial role of mechanical forces in regulating RNA function, paving the way for developing new strategies for RNA manipulation, and potentially advancing molecular engineering. Although the current work is ongoing, we provide current progress of both theoretical and experimental calculations based on our findings.

The purpose of this thesis is to contextualise Hindsight, a sustainability-focused historically based city-simulation and resource management game built by the author. The game and game engine were coded from scratch using the C# programming language and the Unity game development suite of tools. The game focuses on the management of the city of London in two time periods, London from 1850 and the other set in 2050. Both versions of the city are divided into 21 zones, each of which can be managed by the player through the construction, upgrading, or destruction of various buildings within the zone. The player must manage both the city’s resources and the resources of the environment upon which the city depends in order to bring about a more sustainable future and bring the 2050-era version of the city back from the brink of environmental devastation. Along the way, the player must address the cultural views of the society they are managing to ensure their reforms will be accepted and can also see those views slowly change over time. The goal of the game is to provide an interactive learning experience for both the historical element of London and the importance of making sustainable choices.

A significant amount of prior research has been conducted to investigate type 2 diabetes, the most prevalent form afflicting over 90% of diabetic individuals [6]. Yet, gestational diabetes is an understudied form of diabetes that is thought to share various attributes with type 2 diabetes. It was the aim of this project to investigate a proposed mechanism of the disease, the contra-insulin effect, through a cell-culture experiment. To address the question of whether glycemic and hormonal conditions of cell-culture media affect Hs 795.Pl morphology, cellular growth, and glucose uptake, immunocytochemistry (ICC) and a glucose uptake assay was performed. It was hypothesized that higher the presence of hormones, specifically lactogen, in cell culture media will exacerbate the contra-insulin effect, decreasing the glucose uptake of the Hs 795.Pl cells and inducing abhorrent cell morphology. Qualitatively, estradiol and cortisol had a severe impact on cellular morphology indicative of stress and death. As for glucose uptake, it was decreased when the hormones were isolated compared to all together with estradiol thought to be majorly inhibitory to insulin’s proper functioning. It was concluded that cell morphology, growth, and glucose uptake were detrimentally impacted by the gestational hormones, especially those of cortisol and estrogen.

In the United States, nearly every state has a state history museum, where the natural and cultural history of the state is researched and put on display through tangible objects and intangible stories. Despite the unique nature of its history, the state of Hawaii is one of the few states that does not have a proper state history museum. The closest thing the state has to a state history museum is the Bishop Museum, which focuses mostly on Native Hawaiian culture and history from ancient times up until the overthrow of the Hawaiian Kingdom in 1893. This thesis explores why there is no state museum to cover the modern history of the Hawaiian Islands, and lays out a general proposal for what a future Hawaii State History Museum could look like. In researching this project, inspiration was taken from the state history museums of Colorado and Texas, the History Colorado Center and the Bullock Texas State History Museum, respectively. The proposal covers the physical location and design of a potential museum building and how it would be laid out inside. It then provides a list of exhibit ideas while addressing questions of narrative perspective, with the overarching narrative of the museum being “Hawaii History for Hawaii locals.” Lastly, it discusses the importance of the connection between the community and the museum in order to be successful. With current Hawaii legislators showing an interest in actually establishing a state museum for modern Hawaii history, this thesis can serve as a framework from which the real museum can be built off of.
