Reduced space is an important theme in the works of the Marquis de Sade including his epic novel The New Justine and his pornographic performance piece Philosophy in the Bedroom including the political/social treatise "Frenchmen, yet another effort is needed if you want to be a Republic". Through out his life Sade attempted to overcome reduction of space with writing. Tragically, his writing often prolonged the reduction of his space by sending him to or keeping him in prison. It is my theory that his violent, pornographic writing style is "une écriture de surjouissance" or "a writing of over-coming". Surjouissance is my theory for Sade's method, based on textual analysis of Sade's main works, that he combines through his syntactic structure, narrative voice, and semantic themes the orgasm of the mind represented by philosophical discourse with the orgasm of the body represented textually by orgiastic scenes and the language of orgasm to reach an ultimate state of complete freedom. In the political pamphlet "Frenchmen yet another effort..."Sade attempts to set this theory of sur-jouissance, or this transcendent state reached through the combination of physical and philosophical orgasm, as the political foundation for a new republic. Does he succeed in creating a viable political formula for a sustainable republic? My argument states absolutely not. His aristocratic elitism narrows his voice. But he does propose the combination of sexual, literary, and intellectual freedoms as a possible polemic against any form of reduced space.

The video game graphics pipeline has traditionally rendered the scene using a polygonal approach. Advances in modern graphics hardware now allow the rendering of parametric methods. This thesis explores various smooth surface rendering methods that can be integrated into the video game graphics engine. Moving over to parametric or smooth surfaces from the polygonal domain has its share of issues and there is an inherent need to address various rendering bottlenecks that could hamper such a move. The game engine needs to choose an appropriate method based on in-game characteristics of the objects; character and animated objects need more sophisticated methods whereas static objects could use simpler techniques. Scaling the polygon count over various hardware platforms becomes an important factor. Much control is needed over the tessellation levels, either imposed by the hardware limitations or by the application, to be able to adaptively render the mesh without significant loss in performance. This thesis explores several methods that would help game engine developers in making correct design choices by optimally balancing the trade-offs while rendering the scene using smooth surfaces. It proposes a novel technique for adaptive tessellation of triangular meshes that vastly improves speed and tessellation count. It develops an approximate method for rendering Loop subdivision surfaces on tessellation enabled hardware. A taxonomy and evaluation of the methods is provided and a unified rendering system that provides automatic level of detail by switching between the methods is proposed.

Most research on the experience of young women in student service roles in higher education is focused on a reflection of the early career experiences of mid and senior level professionals. Young women enter the field with a set of expectations about the work and their early career experiences need to be uncovered in order to better understand what they expect from their roles in student services. This study focused on the experience of young women in student services and the dynamics they identify as being significant to their work experience. Six women in their mid-twenties working in student service roles participated in two dialogic interviews regarding their work experience. Findings from these women's stories suggest that women are aware of internal and external dynamics that shaped their work experience, and are engaged in their journey toward Self-Authorship along intrapersonal and interpersonal dimensions. Specifically, the women actively chose their career path, looked for opportunities to develop their professional cache, and were impacted by their relationships with their supervisors and colleagues. The women are interested in their professional development in student services in higher education and are active in shaping the experience to meet their expectations. The findings suggest that to understand the experience of young women in student service roles in higher education, women should be asked to share their stories on their early career experiences, including interactions with supervisors and other professional colleagues. By representing these voices in the dialogue on the experience of young women in student service roles, the dynamics that shaped those experiences can be better understood.

Masculinity has been increasingly recognized as a critical and relatively unexplored area of inquiry in anthropological gender studies. This project seeks to expand anthropological research on masculinity to contemporary American society. Using the case study of a male-centered popular new sport, Mixed Martial Arts (also known as cagefighting) this project integrates theories of embodiment and feminist perspectives to explore how masculinity and masculine hegemony are shaped, contested, and perpetuated in the United States. Using a multi-level framework this project explores: 1) How is masculinity experienced and expressed by Mixed Martial Arts fighters as a form of self-identity? How do their bodies play a role in constructing masculinity? 2) What are the pervasive forms of masculinity associated with Mixed Martial Arts (MMA)? Are they truly representative of the sport? 3) Can these pervasive forms of masculinity be seen as hegemonic? How would hegemony operate in relation to individual experience? Using multiple methods to capture multiple points of view was critical to thoroughly examining the complex notion of masculinity. This study employed participant observation, semi-structured interviews, focus groups, surveys, photo elicitation, and media content analysis, as each presented particular benefits and allowed for the development a more well-rounded understanding of masculinity within the realm of MMA. This study also situates the rise of MMA and its representations of masculinity within the greater perspective of contemporary American society. By doing so reveals how ideologies of prescribed masculinity do not arise out of a vacuum but in relation to particular economic, social and political contexts. An emphasis of this study was to examine the daily lives of MMA fighters to understand how their participation in what may be regarded as a hypermasculine activity affects their own perceptions of masculinity. In looking at how masculinity is embodied, the gaps and often contradictions between representation and individual experiences are revealed. Ultimately, the goal of this research is to contribute to a better understanding of masculinity as both an embodied and relational construct.

Since the Convention on Biological Diversity was established in 1992, more importance has been given to the conservation of genetic resources in the international community. In 2001, the International Treaty on Plant Genetic Resources for Food and Agriculture (PGRFA) focused on conserving plant genetic resources, including crop wild relatives (CWR). Some of these genetic resources hold desirable traits--such as transfer of plant disease resistance, improvement of nutritional content, or increased resistance to climate change--that can improve commercial crops. For many years, ex situex situ conservation was the prevalent form of protecting plant genetic resources. However, after PGRFA was published in 1998, in situ techniques have increasingly been applied to conserve wild relatives and enhance domesticated crops.In situ techniques are preferred when possible, since they allow for continued evolution of traits through natural selection, and viability of seed stock through continuous germination and regeneration. In my research, I identified regions in Bolivia and rated them according to their potential for successful programs of iin situ conservation of wild crop relatives. In particular, I analyzed areas according to the following criteria: a) The prevalence of CWRs. b) The impacts of climate change, land use change, population growth, and economic development on the continued viability of CWRs in an area. c) The socio-political and economic conditions that might impede or facilitate successful conservation programs and outcomes. This work focuses on three genera of particular importance in Bolivia: Peanut (Arachis spp.), Potato (Solanum spp.) and Quinoa (Chenopodium spp.). I analyzed the above factors for each municipality in Bolivia (the smallest scale for which appropriate data were available). The results indicate which municipalities are most likely to successfully engage in CWR conservation projects. Finally, I present guidelines for the creation of conservation projects that pinpoint some of the potential risks and difficulties with in situ conservation programs in Bolivia and more generally.

Though some scholars have written about place and history, few have pursued the use of place theory in length in relation to the connections between race, religion, and national identity. Using the writings in the United States and Louisiana in the years surrounding the Louisiana Purchase, I explore place-making and othering processes. U.S. leaders influenced by the Second Great Awakening viewed New Orleans as un-American in its religion and seemingly ambiguous race relations. New Orleanian Catholics viewed the U.S. as an aggressively Protestant place that threatened the stability of the Catholic Church in the Louisiana Territory. Both Americans and New Orleanians constructed the place identities of the other in relation to events in Europe and the Caribbean, demonstrating that places are constructed in relation to one another. In order to elucidate these dynamics, I draw on place theory, literary analysis, and historical anthropology in analyzing the letters of W.C.C. Claiborne, the first U.S. governor of the Louisiana Territory, in conjunction with sermons of prominent Protestant ministers Samuel Hopkins and Jedidiah Morse, a letter written by Ursuline nun Sister Marie Therese de St. Xavior Farjon to Thomas Jefferson, and George Washington Cable's Reconstruction era novel The Grandissimes. All of these parties used the notion of place to create social fact that was bound up with debates about race and anti-Catholic sentiments. Furthermore, their treatments of place demonstrate concerns for creating, or resisting absorption by, a New Republic that was white and Protestant. Place theory proves useful in clarifying how Americans and New Orleanians viewed the Louisiana Purchase as well as the legacy of those ideas. It demonstrates the ways in which the U.S. defined itself in contradistinction to religious others. Limitations arise, however, depending on the types of sources historians use. While official government letters reveal much when put into the context of the trends in American religion at the turn of the nineteenth century, they are not as clearly illuminating as journals and novels. In these genres, authors provide richer detail from which historians can try to reconstruct senses of place.

The term "White flight" and its effects are well documented in large urban city centers. However, few studies consider the same effects on smaller American communities. This case study investigates Lexington, Nebraska, a rural community of approximately 10,000 citizens, that has experienced a population influx of minorities in the last 25 years. The population shift has increased the representation of Hispanic, Asian, and now Somali students in the Lexington Public School system, which, in turn, has been accompanied by a dramatic decrease in White, Anglo students. This study attempts to identify and describe the reasons for the exodus of White students from the public school setting. Possible reasons that might explain the decreases in White student enrollment may include overcrowding in schools, unsafe school environments, and/or less one-on-one attention with classroom teachers.

ATP synthase is a large multimeric protein complex responsible for generating the energy molecule adenosine triphosphate (ATP) in most organisms. The catalysis involves the rotation of a ring of c-subunits, which is driven by the transmembrane electrochemical gradient.

ATP synthase is a large multimeric protein complex responsible for generating the energy molecule adenosine triphosphate (ATP) in most organisms. The catalysis involves the rotation of a ring of c-subunits, which is driven by the transmembrane electrochemical gradient. This dissertation reports how the eukaryotic c-subunit from spinach chloroplast ATP synthase has successfully been expressed in Escherichia coli and purified in mg quantities by incorporating a unique combination of methods. Expression was accomplished using a codon optimized gene for the c-subunit, and it was expressed as an attachment to the larger, more soluble, native maltose binding protein (MBP-c1). The fusion protein MBP-c1 was purified on an affinity column, and the c1 subunit was subsequently severed by protease cleavage in the presence of detergent. Final purification of the monomeric c1 subunit was accomplished using reversed phase column chromatography with ethanol as an eluent. Circular dichroism spectroscopy data showed clear evidence that the purified c-subunit is folded with the native alpha-helical secondary structure. Recent experiments appear to indicate that this monomeric recombinant c-subunit forms an oligomeric ring that is similar to its native tetradecameric form when reconstituted in liposomes. The F-type ATP synthase c-subunit stoichiometry is currently known to vary from 8 to 15 subunits among different organisms. This has a direct influence on the metabolic requirements of the corresponding organism because each c-subunit binds and transports one H+ across the membrane as the ring makes a complete rotation. The c-ring rotation drives rotation of the gamma-subunit, which in turn drives the synthesis of 3 ATP for every complete rotation. The availability of a recombinantly produced c-ring will lead to new experiments which can be designed to investigate the possible factors that determine the variable c-ring stoichiometry and structure.

Telomerase is a specialized enzyme that adds telomeric DNA repeats to the chromosome ends to counterbalance the progressive telomere shortening over cell divisions. It has two essential core components, a catalytic telomerase reverse transcriptase protein (TERT), and a telomerase RNA (TR). TERT synthesizes telomeric DNA by reverse transcribing a short template sequence in TR. Unlike TERT, TR is extremely divergent in size, sequence and structure and has only been identified in three evolutionarily distant groups. The lack of knowledge on TR from important model organisms has been a roadblock for vigorous studies on telomerase regulation. To address this issue, a novel in vitro system combining deep-sequencing and bioinformatics search was developed to discover TR from new phylogenetic groups. The system has been validated by the successful identification of TR from echinoderm purple sea urchin Strongylocentrotus purpuratus. The sea urchin TR (spTR) is the first invertebrate TR that has been identified and can serve as a model for understanding how the vertebrate TR evolved with vertebrate-specific traits. By using phylogenetic comparative analysis, the secondary structure of spTR was determined. The spTR secondary structure reveals unique sea urchin specific structure elements as well as homologous structural features shared by TR from other organisms. This study enhanced the understanding of telomerase mechanism and the evolution of telomerase RNP. The system that was used to identity telomerase RNA can be employed for the discovery of other TR as well as the discovery of novel RNA from other RNP complex.

Many of the works of Dominick Argento have been researched and analyzed, but his choral work Evensong: Of Love and Angels s has received limited attention thus far. Written in memoriam for his wife Carolyn Bailey Argento, Evensong draws its musical material from her initials C.B.A. These letters, translated into note names, form a conspicuous head motive that is present in each movement of the work, and it serves multiple functions: as a melodic feature, as the foundation for a twelve-tone row, and as a harmonic base. This paper provides an overview of the work's conception with specific relation to Argento's biographical details, compositional style, and work habits; a brief review of the critical reception of the work; and a succinct analysis of the form and cyclical materials found in each movement.
